Dual-luciferase assays were performed in the MASMCs cells co-transfected with pRL-TK (Renilla activity for normalization) and the wild-type 2 kb upstream sequence of the Wdr1 gene (Wdr1-WT) or the putative binding site-deletion mutant (Wdr1-MUT) and PCDNA-STAT3 plasmid. ***P < 0.001. (C) ChIP with anti-pSTAT3. MASMCs were serum-starved for 12 h and then treated with IL-6 for 30 min. ChIP assays were performed with a p-STAT3 (Tyr 705) antibody. STAT3 binding to the Wdr1 promoter was examined using PCR with the primers described above (n = 6 per group).
