Fig. 2. Sep15 deficiency arrests cells at the G1 phase by downregulating cyclin E1 and E2 expression. (A?B) Sep15 knockdown caused cell cycle arrest at the G0/G1 phase. After the cell cycle was arrested by the treatment with aphidicolin (A) or nocodazole (B), cell cycle analysis was performed as described in Materials and Methods. The numbers 1 and 2 on the bottom of each panel represent the relative DNA contents of the G0/G1 and G2/M phases, respectively. The number of cells and release time after arrest are marked on the left and right side of Y-axis, respectively. Asy designates asynchronized cells. Data shown are representative of at least three independent experiments. (C) Western blot analyses to measure the levels of p21, p27 and cyclin E1 in shSep15+Dox versus -Dox cells and in shSep15/Rescue+Dox versus -Dox cells.
